The white light laser source of the Leica TCS SP8 X perfectly matches the wavelength of any fluorophore. Up to eight excitation lines can be used – simultaneously.
By tuning both excitation and detection, complete excitation emission spectra can be acquired. With this spectral information, any dye can be optimally excited with minimum cross-excitation and specimen damage.
In addition, the pulsed White Light Laser (WLL) supplies excitation light for fluorescence lifetime measurements. FLIM provides information about the direct environment of a molecule, like local pH or binding to another molecule.

Zero background by LightGateLightGate is a clever, filter-free method to remove unwanted signal from the image data. An adjustable time gate switches off the data collection during the white light laser pulse. Additionally, non-wanted fluorescence can be removed by adjusting the time gate. This way the highest image contrast without any reflection contribution is obtained.

Free selection of FLIM excitation wavelengthThe White Light Laser is an appropriate excitation source for fluorescence lifetime measurements. Its continuous tunability allows performing FLIM measurement at any excitation wavelength. Lifetime of orange and red dyes can now be obtained. The optimal laser repetition rate is selected via an integrated pulse picker.
